One Step FOB Test Cassette

Intended Use

FOB Rapid Test kits and FOB-TRF Rapid test kits, are a fast and visual immune-chromatographic testing devices for the qualitative detection of blood hemoglobin (Fecal Occult Blood, i.e. FOB) in human fecal specimens. These fecal occult blood rapid test kits come in strip and cassette format, and are intended to be used as an aid in the diagnosis of lower gastrointestinal (g.i.) disorders, such as colorectal cancers and adenomas. In this regard, the FOB and FOT-TRF test kits are usually referred to as cancer test device.

Testing Principle

While One-Step FOB-TRF Combo Test is an antigen-capture immunochromatographic assay, detecting the presence of transferrin and fecal occult blood in one single fecal sample. Monoclonal antibodies specifically against human transferrin and hemoglobin, respectively, are 1) conjugated with colloidal gold and deposited on the conjugate pad, and 2) immobilized on the Test Zone (2 and 1) on the nitrocellulose membrane.

When a fecal extraction sample is added, it rehydrates the gold-antibody conjugate and the transferrin and/or hemoglobin, if any in samples, interact with the colloidal gold conjugated antibodies. The antigen-antibody-colloidal gold complex will migrate towards the test window until the Test Zone (2 and 1) where they are captured by immobilized antibodies, forming a visible pink line (indicate positive results). If transferrin and hemoglobin is absent in the sample, no pink line will appear in the Test Zone (2 and 1), indicate negative results.

Why FOB Testing Matters

With our FOB tumor marker rapid test kit, the occult blood can be detected at as low concentration as 40 ng/ml. Since the earlier the diseases like bowel cancer are diagnosed, the better the chance of a cure, an early detection of the FOB in faeces is very important to the treatment of many digestive system disorders. Because usually a cost of a rapid FOB testing kit is almost neglectable, it is highly advisable that the people vulnerable to the related diseases have a screening test regularly.

FOB and TRF: Background information

Fecal occult blood (FOB)refers to the small amount blood in the human faeces, which is not visually detectable. There are several human body disorders which may cause bleeding into the intestines. Ulcers, colitis, polyps, and bowel cancer are the most common ones. In the early stages, cancers in digestive system often cause slight bleeding into the gut, which are often neglected because there are no symptoms at these stages.

Transferrin (TRF) is a blood plasma protein for iron ion delivery. A patient with an increased serum transferrin level often suffers from iron deficiency anemia that is one of the most important symptoms when a tumor has caused chronic occult bleeding.

  • Single Sample Collection Tube with solution;
  • Instruction Insert.

Specimen

Faeces Preparation

Collect feces into a clean, dry specimen collection container.

Unscrew the cap of the specimen collection tube, and randomly poke the specimen collection stick fixed in the cap into the fecal specimen on at least 3 different sites. Do not scoop the fecal specimen.

Tighten the cap of the specimen collection tube, and shake the tube vigorously to mix the specimen and the extraction buffer.

The mixed liquid is the specimen that can be tested immediately.

Best results will be obtained if the assay is performed within 6 hours after collection.

Specimen prepared in the specimen collection tube may be stored for 6 months at -20°C.

Let the specimen come to room temperature naturally before testing the refrigerated specimens.

In the test band region, antibody specific to the target antigen or protein is immobilized. In the end of the membrane strip where the samples are added, another antibody conjugated with colloidal gold particles is embedded.

During the testing process, specimen from the patient is added to the end of the membrane strip. The antibody conjugate particles will resolve into the specimen, where the antibody conjugate will react and bound to the target protein or viral antigen, if present.

This antigen-antibody conjugate complex then moves along the strip to the testing band region, under capillary effect. The immobilized antibody in the test line region then bound to another binding site on the antigen complex, and the antibody-antigen-antibody complex is fixed on test band region.

When the complex congregates, the highly concentrated colloidal gold particles will develop some red or pink color, indicate a positive result.

If there is no target protein or antigen in the specimen, the colloidal gold particles will not congregate and no color will develop, indicating a negative result.
